Composition

Inj: Per 1 ml amp: Prochlorperazine mesilate 12. 5 mg.Tab 3 mg/5 mg: Prochlorperazine maleate 3 mg/5 mg.

Indications & Dosage

Nausea and vomiting, acute attack: ORAL: Adult: Initially 20 mg, then 10 mg after 2 hours BY DEEP IM INJ: Adult: 12.5 mg as required, followed by an oral dose after 6 hours,if necessary. Nausea and vomiting, prevention and treatment: ORAL: Adult: 5-10 mg 2-3 times a day. Children 1-11 years (body-weight 10 kg and above): 250 mcg/kg 2-3 times a day; 12-17 years: 5-10 mg up to 3 times a day ifrequired. BY DEEP IM INJ: Adult: 12.5 mg as required, followed by an oral dose after 6 hours, if necessary. Children 2-4 years: 1.25-2.5 mg up to 3 times a day if required; 5-11 years: 5-6.25 mg up to 3 times a day if required; 12-17 years: 12.5 mg up to 3 times a day if required Labyrinthine disorders: ORAL: Adult: 5 mg 3 times a day, increased if necessary to 30 mg daily in divided doses, increased gradually, then reduced to 5-10 mg daily, dose is reduced after several wks Nausea and vomiting in previously diagnosed migraine: ORAL: Adult & Children 12 yrs: 3-6 mg twice daily. Schizophrenia and other psychoses, Mania: ORAL: Adult: 12.5 mg twice daily for 7 days, dose to be adjusted at intervals of 4-7 days according to response; usual dose 75-100 mg daily. BY DEEP IM INJ: Adult: 12.5-25 mg 2-3 times a day. Short-term adjunctive management of severe anxiety: ORAL: Adult: 15-20 mg daily in divided doses; max. 40 mg per day. Doses are expressed as prochlorperazine maleate or mesilate; 1mg prochlorperazine maleate:1 mg prochlorperazine mesilate.

Administration

IV: Administer IV doses as 30 min infusion or by slow (5-10 min) IV push. IV infusion must be diluted in 50-100 ml NS or D5W. IV injection may be given undiluted. IM: Inject IM form slowly, deep into upper outer quadrant of buttocks. Keep patient supine for 30 mins after injection to minimize hypotensive effects. Rotate IM Inj sites to prevent irritation and sterile abscesses.

Compatibility

Stable in NS, D5W, D1 OW, LR, 1/2NS, DSNS, DSLR.

Stability

Intact vials or ampoules are stable at room temperature. Protect from light.

Contraindications

Children under 10 kg body weight, CNS depression, comatose states, phaeochromocytoma, bone marrow depression, liver impairment.

Precautions

Undiagnosed and prolonged vomiting, elderly, hypotension, cardiovascular disease.

Interactions

CNS depressants, alcohol, analgesics, antihypertensives, antidepressants, anticonvulsants, antidiabetics.

Side Effects

Impaired judgment and dexterity, CNS disturbances, acute dys tonic reactions such as facial and skeletal muscle spasms and oculogyric crises, anticholinergic effects, ECG and endocrine changes, allergic effects and extra-pyramidal symptoms occur rarely at low dosage.
